چکیده
Visceral Leishmaniasis (VL) is a disease of poor and neglected populations; it affects 79 countries of the world and accounts 58,000 new cases to each year. However, no effective treatment for cure of the disease is yet available. Therefore, the present study was designed to assess the curative efficacy of single dose of sodium stibogluconate in combination with Killed Leishmania donovani (KLD) antigen/78kDa antigen along with MPL-A in murine visceral leishmaniasis. After 30 days infection, infected mice were treated with a single dose of chemotherapy or immunochemotherapy. These animals were then sacrificed after 30 days post treatment and determined for hepatic parasite load and their immunological profile. It was observed that animals treated with a combination of SSG and immunotherapy not only reduced the parasite load but the immune profile was shifted to protective Th1 type (elevated levels of IFN-gamma and IL-2 were observed) of immune response. Hence our study lay emphasis on the use of short course (single dose) combination therapy of SSG and 78kDa/KLD along with MPL-A.

Immunization of C57BL/6 Mice with GRA2 Combined with MPL Conferred Partial Immune Protection against Toxoplasma gondii
BBackground: We have previously reported that immunization with GRA 2 antigen of Toxoplasma gondii induces protective immunity in CBA /J (H2k) and BALB/c mice (H2d). We aimed to examine whether immunization of a distinct strain of rodent with recombinant dense granule antigens (GRA2) combined with monophosphorryl lipid A (MPL) adjuvant elicits protective immune response against T. gondii. Metho...
متن کاملTherapy with Sodium Stibogluconate in Stearylamine-Bearing Liposomes Confers Cure against SSG-Resistant Leishmania donovani in BALB/c Mice
BACKGROUND Resistance of Leishmania donovani to pentavalent antimonials, the first-line treatment of visceral leishmaniasis (VL), has become a critical issue worldwide. Second-line and new drugs are also not devoid of limitations. Suitable drug-delivery systems can improve the mode of administration and action of the existing antimonials, thus increasing their clinical life. METHODOLOGY/PRINC...
متن کاملMorphological Findings in Trophozoites during Amoebic Abscess Development in Misoprostol-Treated BALB/c Mice
During amoebic liver abscess (ALA) formation in susceptible animals, immune response is regulated by prostaglandin E2 (PGE2) dependent mechanisms. The aim of this study was to analyze the effect of misoprostol (MPL), a PGE1 analogue, on ALA formation in BALB/c mice. Male mice from BALB/c strain were intrahepatically infected with 7.5 × 10(5) trophozoites of E. histolytica strain HM1:IMSS and tr...
متن کاملStudy on immunity of Leishmania major new crude antigen as a vaccine against leishmaniasis in out bred (resistant) and Balb/c (sensitive) mice
The aim of this study was to prepare a new formula of Leishmania major (L. major) crude antigen and evaluate its effect on immune system. For this purpose L. major promastigotes were cultured, harvested, washed, and resuspended in physiologic saline and the suspensions were dispersed in five equal batches. 0.1 ml of various doses of the cocktail antigen were injected intradermaly in three group...
